目的:探讨脑泰方提取物对大鼠局灶性脑缺血后血管内皮生长因子(VEGF)及VEGF mRNA的影响。方法:30只SD大鼠随机分为假手术组、模型组、脑复康组、脑泰方低、中、高剂量组。通过月桂酸钠损伤血管内皮细胞来复制局灶性脑缺血模型,采用免疫组化和RT-PCR法检测各组大鼠脑内VEGF阳性细胞数和VEGF mRNA的表达,同时评价大鼠神经功能。结果:脑缺血后,药物组神经功能症状得到改善,尤以脑泰方中剂量组变化最显著(P<0.01);假手术组大鼠脑内有少量VEGF阳性细胞,脑缺血后VEGF阳性细胞表达增加,与模型组比较,脑泰方中剂量组VEGF阳性细胞数明显增多(P<0.01),而且VEGF mRNA的表达也显著加强(P<0.01)。结论:脑泰方提取物增强大鼠局灶性脑缺血后VEGF及VEGF mRNA的表达,可能是其抗脑缺血的作用机制之一。
Objective: To investigate the effect of Naotaifang extract on vascular endothelial growth factor (VEGF) and VEGF mRNA after focal cerebral ischemia in rats. METHODS: Thirty SD rats were randomly divided into sham operation group, model group, Naofukang group, and Naotai low, middle, and high dose groups. The vascular endothelial cells were injured by sodium laurate to induce focal cerebral ischemia. Immunohistochemistry and RT-PCR were used to detect the expression of VEGF-positive cells and VEGF mRNA in rat brain. . RESULTS: After cerebral ischemia, the neurological symptoms of the drug group improved, especially in the middle-dose group of Naotaifang (P<0.01). There was a small amount of VEGF-positive cells in the brain of sham-operated rats, and VEGF after cerebral ischemia. The expression of positive cells was increased. Compared with the model group, the number of VEGF-positive cells in the middle-dose group was significantly increased (P<0.01), and the expression of VEGF mRNA was also significantly increased (P<0.01). Conclusion: Naotaifang extract enhances the expression of VEGF and VEGF mRNA after focal cerebral ischemia in rats, which may be one of the mechanisms of the anti-cerebral ischemia.
