目的观察补肾益智方对AD模型大鼠脑内Ch AT和Ach E水平的影响及可能的量效关系。方法将Wistar大鼠随机分为5组,并制备AD大鼠模型,应用放射性化学法和微量羟胺比色法分别对大鼠脑内组织的Ch AT(前额皮质、中隔、海马)、Ach E(前额皮质、海马)活性进行检测,并设立石杉碱甲组和正常老年组进行对比研究。结果与模型组比较,补肾益智方两个组对前额皮质、海马两个酶的活性均有不同程度的调节作用,尤其低剂量组效果为优。说明补肾益智方可明显改善该模型的胆碱能功能低下状态,且存在着一定的量效关系,其中低剂量组治疗效果优于石杉碱甲组。结论研究提示本方对AD模型大鼠海马Ch AT和Ach E的活性的调节作用,可能是其治疗AD作用的机制之一。
Objective To observe the effects of Bushen Yizhi Decoction on the levels of Ch AT and Ach E in brain of AD model rats and its possible dose-effect relationship. Methods Wistar rats were randomly divided into 5 groups and AD model was established. ChAT (prefrontal cortex, septum, hippocampus), Ach E (Prefrontal cortex, hippocampus) activity were detected, and the establishment of huperzine A group and normal elderly group comparative study. Results Compared with the model group, Bushen Yizhi Decoction could regulate the activity of the two enzymes in the prefrontal cortex and hippocampus to varying degrees, especially in the low dose group. The results showed that Bushen Yizhi could significantly improve the model of cholinergic dysfunction, and there is a certain dose-effect relationship, in which low-dose group is better than huperzine A group. Conclusions This study suggests that this prescription regulates the activity of ChAT and Ach E in hippocampus of AD model rats, which may be one of the mechanisms of AD treatment.
