目的 :探讨肝细胞生长因子(hepatocyte growth factor,HGF)对胃癌细胞株AGS增殖、迁移和侵袭的影响。方法:体外培养AGS细胞,应用浓度为25、50、100 ng/m L的HGF处理AGS细胞,采用CCK-8法检测AGS细胞的增殖,transwell法分别检测HGF对AGS细胞迁移、侵袭能力的影响。结果:CCK-8法检测结果显示,HGF对AGS细胞有促增殖作用。随着HGF浓度的增加或作用时间的延长,细胞增殖活力呈上升趋势,在72 h最为明显,呈时间、浓度依赖关系。Transwell迁移、侵袭实验显示,不同浓度的HGF对AGS细胞的迁移和侵袭能力有促进作用,以50 ng/m L最为明显(P<0.01)。结论 :HGF具有促进胃癌细胞增殖、迁移和侵袭的作用。
Objective: To investigate the effect of hepatocyte growth factor (HGF) on proliferation, migration and invasion of gastric cancer cell line AGS. Methods: AGS cells were cultured in vitro. AGS cells were treated with 25, 50 and 100 ng / mL HGF. The proliferation of AGS cells was detected by CCK-8 assay. The effect of HGF on the migration and invasion of AGS cells was detected by transwell assay . Results: The results of CCK-8 assay showed that HGF promoted the proliferation of AGS cells. With the increase of HGF concentration or the prolongation of action time, the cell proliferation activity showed an upward trend, which was the most obvious at 72 h in a time and concentration dependent manner. Transwell migration and invasion experiments showed that different concentrations of HGF promoted the migration and invasion ability of AGS cells, and the most obvious was 50 ng / mL (P <0.01). Conclusion: HGF can promote the proliferation, migration and invasion of gastric cancer cells.
