目的探讨PITX1和突变型P53基因在胃癌发生发展过程中的作用。方法采用RT-PCR和免疫组织化学方法联合检测43例正常胃组织、27例肠上皮化生、19例不典型增生及22例胃癌中PITX1和突变型p53 mRNA和蛋白的表达。结果在正常胃组织,肠上皮化生,不典型增生和胃癌中PITX1的表达逐渐降低,胃癌组与其他各组相比差异有统计学意义(均P<0.05),而突变型P53表达逐渐升高,胃癌组与正常组和肠上皮化生组相比差异显著(均P<0.05),与不典型增生组相比差异无统计学意义(P>0.05)。结论 PITX1与P53两种分子的异常表达在胃癌的发生发展中可能具有协同作用,二者联合检测能为胃癌的早期临床诊断和治疗提供有利的生物学信息。
Objective To investigate the role of PITX1 and mutant P53 genes in the development of gastric cancer. Methods RT-PCR and immunohistochemical methods were used to detect the expression of PITX1 and mutant p53 mRNA and protein in 43 cases of normal gastric tissue, 27 cases of intestinal metaplasia, 19 cases of atypical hyperplasia, and 22 cases of gastric cancer. Results The expression of PITX1 gradually decreased in normal gastric tissue, intestinal metaplasia, atypical hyperplasia, and gastric cancer. The difference between gastric cancer group and other groups was statistically significant (all P<0.05), while that of mutant P53 gradually increased. There was significant difference between the high and the gastric cancer groups compared with the normal group and the intestinal metaplasia group (all P<0.05). There was no significant difference between the gastric cancer group and the atypical hyperplasia group (P>0.05). Conclusion The abnormal expression of PITX1 and P53 may play a synergistic role in the occurrence and development of gastric cancer. The combination of the two may provide favorable biological information for the early clinical diagnosis and treatment of gastric cancer.
