目的 探讨肾脏特异性表达新基因Collectrin在大鼠 5 / 6肾切除残余肾组织中的表达变化 ,为Collectrin基因功能研究提供线索。方法 建立慢性肾衰大鼠 5 / 6肾切除模型 ,分为对照组( 10只 )、手术组 ( 12只 )和依那普利治疗组 ( 12只 ) ,采用逆转录 PCR和Western蛋白印迹、免疫组织化学技术检测手术后 2、8和 16周大鼠肾组织Collectrin的表达变化。结果 在对照组中 ,随着大鼠周龄的增长大鼠CollectrinmRNA表达逐渐减少。 2 ,8,16周分别为 ,1 6 9± 0 0 9,1 38± 0 0 4,1 2 1± 0 0 9,(P <0 0 5 ) ;在术后 2 ,8,16周 ,手术组 ( 2 33± 0 0 7,1 85± 0 2 0 ,1 47± 0 17)Collectrin表达明显高于对照组 ( 1 6 9± 0 0 9,1 38± 0 0 4,1 2 1± 0 0 9,P <0 0 5 ) ;经依那普利治疗后 ,Collectrin表达明显减少 ;手术组Collectrin的表达在手术后 2周明显增加 ,随着慢性肾衰的进展Collectrin的表达逐渐减少。 2 ,8,16周分别为 2 33± 0 0 7,1 85± 0 2 0 ,1 47± 0 17(P <0 0 5 )。Collectrin蛋白的表达情况与CollectrinmRNA表达情况相一致。结论 Collectrin的表达随大鼠周龄的增长而减少 ;在 5 / 6肾切除残余肾组织中 ,Collectrin的表达与肾脏的增生、肥大的病生理变化相关。
Objective To investigate the expression of Collectrin, a novel gene specific for kidney, in residual renal tissue in 5/6 nephrectomy in rats, and provide clues for the study of the gene function of Collectrin. Methods A 5/6 nephrectomy model was established in rats with chronic renal failure. The model was divided into control group (n = 10), operation group (n = 12) and enalapril treatment group (n = 12). Reverse transcription PCR and Western blotting were used. Immunohistochemistry was used to detect the changes of Collectrin expression in renal tissues at 2, 8 and 16 weeks after operation. Results In the control group, the expression of Collectrin mRNA gradually decreased with the increase of the rat’s age. 2, 8 and 16 weeks, respectively, was 169 ± 0 0 9,1 38 ± 0 0 4,1 2 1 ± 0 0 9 (P 0 05). At 2, 8 and 16 weeks after operation, The expression of Collectrin in the operation group was significantly higher than that in the control group (2 33 ± 0 0 1, 1 85 ± 0 2 0, 1 47 ± 0 17) (169 ± 0 0 9,1 38 ± 0 0 4,1 2 1 ± The expression of Collectrin decreased significantly after enalapril treatment. The expression of Collectrin in the operation group increased significantly 2 weeks after operation, and the expression of Collectrin decreased gradually with the progression of chronic renal failure. 2, 8 and 16 weeks were respectively 2 33 ± 0 0 7,1 85 ± 0 2 0,1 47 ± 0 17 (P 0 05). Collectrin protein expression and Collectrin mRNA expression consistent. Conclusions The expression of Collectrin decreases with the growth of the rat ’s age. In 5/6 nephrectomized residual kidney tissue, the expression of Collectrin is related to the changes of renal hyperplasia and hypertrophy.
