目的分析新疆乌鲁木齐市2008年成人麻疹爆发流行的麻疹野病毒的基因特征。方法用Vero/slam细胞从成人麻疹暴发患者的标本中分离出麻疹野病毒,应用逆转录一聚合酶链反应(RT-PCR)对麻疹野病毒分离株扩增核蛋白N基因羧基末端676个核苷酸片段,再对扩增产物进行核苷酸序列测定和分析。并以N基因羧基末端450个核苷酸片段构建基因亲缘性关系树,进行核苷酸变异分析。结果首批共分离到5株麻疹野病毒,这些病毒N基因450个核苷酸之间的差异为0%~0.2%,均为H1基因型中的Hla基因亚型。结论引起新疆乌鲁木齐市2008年成人麻疹流行的麻疹野病毒为Hla基因亚型。
Objective To analyze the genetic characteristics of wild measles virus in 2008 with outbreak of adult measles in Urumqi, Xinjiang. Methods Vero / slam cells were isolated from specimens of adult measles outbreaks and wild-type measles virus was amplified by RT-PCR from 676 nucleotides Nucleotide fragment, and then the amplified product was subjected to nucleotide sequence determination and analysis. Nucleotide sequence analysis was carried out by constructing a phylogenetic tree of 450 nucleotides at the carboxyl terminus of N gene. Results Five strains of wild measles virus were isolated in the first batch. The nucleotide variation of 450 nucleotides in these N genes was 0% -0.2%, all of which were Hla subtypes in H1 genotype. Conclusion The measles wild virus which caused the epidemic of adult measles in Urumqi, Xinjiang in 2008 was Hla subtype.
