目的 了解高三尖杉酯碱 (HHT)诱导CNE 2Z细胞凋亡过程中Caspase 6、 8的活性变化。方法 用不同浓度 (0 .12 5、0 .2 5、0 .5、1μg/ml)的HHT分别处理CNE 2Z细胞 1、2、4、8、12、2 4h ,用比色法检测其Caspase 6、 8的活性 (A40 5nm表示 )。结果 1μg/mlHHT处理CNE 2Z细胞 1h即可见Caspase 8活性明显高于两个对照组 (未处理组和抑制剂 +药物处理组 ,P <0 .0 1) ,2h达高峰。而Caspase 6的活性在 4h开始升高 ,12h达高峰 ,2 4h仍明显高于对照组 (P <0 .0 1)。不同浓度的HHT处理CNE 2Z细胞 ,Caspase 6在 12h、Caspase 8在 2h时均可见它们的活性明显高于两个对照组 (P <0 .0 1) ,且随HHT浓度的增加而活性增强。结论 Caspase 6、 8参与了HHT诱导的CNE 2Z细胞凋亡 ,且其活性升高有时间和浓度依赖性。
Objective To investigate the changes of Caspase 6, 8 activity in homoharringtonine (HHT) -induced CNE 2Z cell apoptosis. Methods CNE 2Z cells were treated with HHT at different concentrations (0.12 5,0 .2 5,0. 5 and 1 μg / ml) for 1, 2, 4, 8, 12 and 24 hours respectively. Caspase 6, 8 activity (A40 5nm representation). Results Caspase 8 activity in CNE 2Z cells treated with 1μg / ml HHT for 1h showed that the activity of Caspase 8 was significantly higher than that of the two control groups (untreated and inhibitor + drug treated groups, P <0.01) and peaked at 2h. The activity of Caspase 6 began to rise at 4h, peaked at 12h, and remained significantly higher at 24 h than that of the control group (P <0.01). The CNE2Z cells treated with different concentrations of HHT showed that Caspase 6 at 12h and Caspase 8 at 2h showed significantly higher activities than those of the two control groups (P <0.01), and increased with the increase of HHT concentration. Conclusion Caspase 6 and 8 are involved in HHT-induced apoptosis of CNE 2Z cells, and their activities are increased in a time and concentration-dependent manner.
