目的在人参属资源中研究20(R)-25-羟基-原人参三醇[20(R)-25-OH-PPT],为人参皂苷降血糖作用的创新和研发提供科学依据。方法色谱柱Agilent C18(250 mm×4.6 mm,5μm);流动相为甲醇-水(70∶30);柱温35℃;体积流量1.0 m L/min;ELSD气化室温度87.0℃,载气流速2.20 L/min;进样体积10μL。采用HPLC-ELSD法测定人参属植物不同部位提取物(以丙酮为溶剂,采用超声法提取)中20(R)-25-OH-PPT。结果 20(R)-25-OH-PPT在0.001~0.050 mg/m L线性关系良好,在人参茎叶、人参根、人参果、人参花、西洋参茎叶、西洋参根和三七芦头总皂苷中的量分别为3.74%、2.59%、2.37%、0.40%、0.68%、0.22%和0.39%,回收率为98.54%、97.54%、100.35%、101.46%、97.46%、99.14%和99.38%。结论20(R)-25-OH-PPT存在于人参属植物多个部位中,可作为人参皂苷降血糖作用开发利用的重要依据。
Objective To study the 20 (R) -25-hydroxy-protopanaxatriol [20 (R) -25-OH-PPT] in ginseng resources and provide a scientific basis for the innovation and research on the hypoglycemic effect of ginsenosides. Methods The column was Agilent C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was methanol-water (70:30). The column temperature was 35 ℃. The volume flow rate was 1.0 m L / min. Speed 2.20 L / min; injection volume 10 μL. The content of 20 (R) -25-OH-PPT in extracts from different parts of Panax ginseng (extracted with acetone and extracted by ultrasonic method) was determined by HPLC-ELSD. Results 20 (R) -25-OH-PPT had a good linearity in the range of 0.001-0.050 mg / m L. In the stems and leaves of ginseng, ginseng root, ginseng fruit, ginseng flower, stem and leaf of American ginseng, The recoveries were 98.54%, 97.54%, 100.35%, 101.46%, 97.46%, 99.14% and 99.38%, respectively. The recoveries were 3.74%, 2.59%, 2.37%, 0.40%, 0.68%, 0.22% and 0.39%, respectively. Conclusion 20 (R) -25-OH-PPT exists in many parts of Panax ginseng and can be used as an important basis for the development and utilization of ginsenosides.
