目的探讨康心汤对腹主动脉缩窄心肌肥厚大鼠模型心脏Ⅰ型胶原、Ⅲ型胶原mRNA表达的作用。方法造模4周后,予以康心汤干预,实验分假手术组、模型组、模型组+康心汤大、中、小剂量组。假手术组(生理盐水),模型组(生理盐水),模型+康心汤低剂量组[2.5g/(kg·d)],模型+康心汤中剂量组[5g/(kg·d)],模型+康心汤高剂量[10 g/(kg·d)]灌胃,药物干预8周后无痛处死大鼠取材,测定左心室质量指数,RT-PCR检测心脏Ⅰ、Ⅲ型胶原mRNA。结果模型组左心室重量增加,左心室质量指数增加,心脏Ⅰ、Ⅲ型胶原mRNA表达上调,康心汤降低左心室质量指数,下调心脏Ⅰ、Ⅲ型胶原mRNA表达。结论康心汤可以调节心脏Ⅰ、Ⅲ型胶原mRNA表达缓解腹主动脉缩窄大鼠模型心肌肥厚和纤维化。
Objective To investigate the effect of Kangxin Decoction on the expression of type Ⅰ collagen and type Ⅲ collagen mRNA in heart of hypertrophic rat with abdominal aorta constriction. Methods 4 weeks after the model was given Kangxin Tang intervention, the experimental sub-surgery group, model group, model group + Kang Xin Tang large, medium and small dose group. Model group (saline), low dose Kangxin Decoction group (2.5g / (kg · d)], model group + Kangxin Tang middle dose group [5g / (kg · d) ], Model + Kang Xin Tang high dose [10 g / (kg · d)] intragastric administration, the rats were painlessly killed after 8 weeks of drug intervention, the left ventricular mass index was measured, RT-PCR detection of cardiac collagen type Ⅰ, Ⅲ mRNA. Results In the model group, the left ventricular mass increased, the left ventricular mass index increased, and the expressions of type Ⅰ and type Ⅲ collagen mRNA increased. Kangxin Tang decreased the left ventricular mass index and decreased the expression of type Ⅰ and type Ⅲ collagen mRNA. Conclusion Kangxin Decoction can regulate the expression of collagen type Ⅰ and type Ⅲ mRNA in cardiac hypertrophy and fibrosis in rats with abdominal aorta stenosis.
