目的 探讨大鼠睾丸组织在短期糖尿病状态下氧自由基和抗氧化水平的变化。方法 4 8只Wistar雄性大鼠被随机分为 6组 ,每组 8只 :其中 1组腹腔注射枸橼酸缓冲液作为正常对照组 ,其余 5组腹腔注射链脲菌素 ,分别观察 1,3,5 ,7,14d。观察期满后断头处死 ,取睾丸 ,制备组织匀浆 ,分别检测组织中SOD、MDA、GSH、GST、GSH px、NO和NOS水平。结果 与正常大鼠相比 ,糖尿病大鼠睾丸组织中总SOD活力下降 ,MDA含量上升 ;GSH含量和GSH px 活力无显著性变化 ,GST活力下降 ;NO含量及NOS活力均下降。结论 睾丸组织的氧自由基 ,主要是O2 含量增加以及NO合成受抑制 ,可能与糖尿病大鼠性与生殖功能障碍有关
Objective To investigate the changes of oxygen free radicals and antioxidation in rat testis under short-term diabetic conditions. Methods Forty eight Wistar male rats were randomly divided into 6 groups with 8 rats in each group: 1 group was intraperitoneally injected with citrate buffer as normal control group, and the other 5 groups were injected intraperitoneally with streptozotocin to observe 1,3 , 5, 7, 14d. After the observation period, the rats were decapitated and the testes were taken to prepare the tissue homogenates. The levels of SOD, MDA, GSH, GST, GSH px, NO and NOS in the tissues were measured respectively. Results Compared with normal rats, the activity of total SOD in testis tissue of diabetic rats decreased and the content of MDA increased. The content of GSH and the activity of GSH px did not change significantly but the activity of GST decreased. The content of NO and the activity of NOS decreased. Conclusion The testicular tissue oxygen free radicals, mainly O2 content increased and NO synthesis inhibited, may be related to sexual and reproductive dysfunction in diabetic rats
