目的探讨脾酪氨酸激酶(Syk)在胃癌组织中的表达及其对细胞凋亡的影响。方法采用免疫组化SP法,检测68例胃癌组织及其癌旁正常组织中Syk蛋白表达情况,流式细胞术检测胃癌组织中肿瘤细胞凋亡情况。结果胃癌组织中Syk蛋白阳性表达率低于癌旁正常组织(20.59%vs.83.82%)(P<0.05)。淋巴结转移(41例)的胃癌组织中Syk蛋白阳性表达率和细胞凋亡率均低于无淋巴结转移(27例)的胃癌组织[(2.44%vs.48.15%和(2.95±1.56)%vs.(16.82±11.70)%)](P<0.05)。Syk蛋白阳性表达(14例)的胃癌组织细胞凋亡率高于Syk蛋白阴性表达(54例)的胃癌组织[(26.68±7.31)%vs.(3.73±2.11)%](P<0.05)。结论 Syk蛋白可通过诱导肿瘤细胞凋亡对胃癌细胞的生长和转移有抑制作用。
Objective To investigate the expression of Spleen tyrosine kinase (Syk) in gastric carcinoma and its effect on apoptosis. Methods Immunohistochemical SP method was used to detect the expression of Syk protein in 68 cases of gastric cancer and adjacent normal tissues. Flow cytometry was used to detect the apoptosis of tumor cells in gastric cancer. Results The positive rate of Syk protein expression in gastric cancer tissues was lower than that in adjacent normal tissues (20.59% vs.83.82%, P <0.05). The positive rates of Syk protein and apoptosis in gastric cancer with lymph node metastasis (41 cases) were lower than those without lymph node metastasis (27 cases) [(2.44% vs.48.15% vs (2.95 ± 1.56)% vs. (16.82 ± 11.70)%)] (P <0.05). The apoptosis rate of Syk-positive (14 cases) gastric cancer tissue was higher than that of Syk-negative (54 cases) (26.68 ± 7.31)% vs 3.73 ± 2.11% (P <0.05). Conclusion Syk protein can inhibit the growth and metastasis of gastric cancer cells by inducing tumor cell apoptosis.
