目的:比较不明原因复发性流产(URSA)与正常早孕蜕膜CD8和颗粒酶B的表达,从蛋白水平探讨不明原因复发性流产的发病机制。方法:用免疫组织化学方法测定两组蜕膜CD8和颗粒酶B蛋白表达,采用双盲法随机取5个视野,400倍下观察蜕膜组织处阳性细胞,计数后取其平均数进行半定量分析。结果:与正常早孕相比,URSA组蜕膜组织处颗粒酶B表达明显升高(P<0.01);CD8的表达两组比较无明显差异(P>0.05)。结论:早孕期蜕膜组织颗粒酶B蛋白表达异常是URSA的机制之一;颗粒酶B死亡途径可能是早期诱导URSA发生的重要因素。
Objective: To compare the expression of CD8 and granzyme B in unexplained recurrent spontaneous abortion (URSA) and normal early pregnancy deciduals and to explore the pathogenesis of unexplained recurrent spontaneous abortion from the protein level. Methods: The expressions of CD8 and granzyme B in decidua were determined by immunohistochemical method. Five fields were randomly selected by double-blind method and the number of positive cells in decidual tissue was observed under 400 times. analysis. Results: Compared with normal early pregnancy, the expression of granzyme B in decidual tissue of URSA group was significantly increased (P <0.01). There was no significant difference between the two groups in the expression of CD8 (P> 0.05). CONCLUSIONS: The abnormal expression of granzyme B in early decidual tissue is one of the mechanisms of URSA. The pathway of granzyme B death may be an important factor in the early induction of URSA.
