目的:探讨维生素D(VitD)对分化中期的骨骼肌细胞c2c12的胰岛素信号通路的影响。方法用VitD10 nm或是100 nm刺激分化4 d的骨骼肌细胞c2c121 d,细胞回收前进行或不进行5 min的50 nm胰岛素的刺激。Western印迹法测定胰岛素信号通路中细胞外信号调节激酶(ERK)和丝氨酸/苏氨酸蛋白激酶(AKT)的磷酸化水平。结果 VitD单独处置能够提高ERK的磷酸化水平,并且呈剂量依赖性。其与胰岛素联合处置引起的ERK磷酸化为加和效应;VitD单独处置没有改变AKT的磷酸化水平,与胰岛素联合应用也没有改变胰岛素引起的AKT的磷酸化水平。结论在分化中期的骨骼肌细胞c2c12中,VitD通过加和效应提高了胰岛素信号通路中ERK的磷酸化,但没有影响AKT的磷酸化水平。“,”Objective To explore the effect of vitamin D (VitD) on insulin signal pathway in differentiating myoblast c2c12 cell. Methods Differentiating myoblast c2c12 cells were stimulated 1 day with VitD10nM or 100nM. Before sampled,cells were administrated with or without 50nM insulin 5 minutes. Phosphorylation levels of ERK and AKT in the insulin signaling pathway were detected by Western blotting. Results Phosphorylation level of ERK was enhanced in cells treated with only VitD. And the increase showed dose-dependent. Insulin also singly induced high phosphorylation level of ERK. phosphorylation level of ERK treated with the combination of VitD and insulin was higher than that with single each treatment. The single VitD administration could not enhanced phosphorylation level of AKT. VitD could not further increase the phosphorylation of AKT induced by insulin. Conclusion VitD did not cause phosphorylation of AKT, but caused phosphorylation of ERK in differentiating myoblast c2c12. The phosphorylation of ERK caused by the combination of VitD and insulin was additive effect.
