血管平滑肌细胞(VSMC)异常增殖是动脉粥样硬化等疾病的主要病理过程之,抗VSMC增殖就可有效地防治动脉粥样硬化。本实验选择陈淑长教授临床治疗早期肢体动脉硬化性闭塞症(ASO)的有效方药温脉通,观察其对VSMC增殖及细胞周期的影响,同时测定培养液中SOD活性、脂质过氧化物的含量,探讨该方抑制VSMC增殖、防治ASO的作用机理。1实验材料1.1药品与试剂温脉通提取液由北京中医药大学东方医院制剂中心提供,经水煮、醇沉后制成提取液(每毫升含中药生药为3.046g)。AngⅡ、EDTA、MTT、PI、RNA酶(美国SIGMA公司),胎牛血清、DMEM、胰蛋白
The abnormal proliferation of vascular smooth muscle cells (VSMC) is the main pathological process of atherosclerosis and other diseases. Anti-VSMC proliferation can effectively prevent atherosclerosis. In this study, Professor Chen Shuchang chose Wenmaitong, an effective medicine for the treatment of early limb arteriosclerosis obliterans (ASO), to observe its effect on VSMC proliferation and cell cycle, and to measure SOD activity and lipid peroxide content in the culture fluid. To investigate the mechanism of the inhibition of VSMC proliferation and the prevention and treatment of ASO. 1 Experimental Materials 1.1 Drugs and Reagents Wenmaitong extract was provided by the preparation center of Oriental Medicine Hospital of Beijing University of Traditional Chinese Medicine.
