目的探讨扶正祛毒法延缓肝纤维化进展的最佳给药时机。方法采用CCl4皮下注射12周的方法制作大鼠肝纤维化模型,中药组给予芪莲纤化汤治疗,并依据给药时间不同分为造模第1周(芪莲1组)、第4周(芪莲2组)、第7周(芪莲3组)3个给药组,西药治疗组(DDB组)从第1周的第1天开始给予联苯双酯滴丸治疗,观察不同组别、不同时期的血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、血清白蛋白(ALB)3项肝脏功能生化指标和肝组织病理。结果注射CCl4后,模型组大鼠一般状况逐渐变差,体重较正常组减轻,后期有腹水出现;第6周开始转氨酶显著升高,第9周末血清白蛋白明显下降;在第6周后出现明显的纤维间隔,肝小叶被破坏、紊乱,9周末肝内假小叶广泛形成。芪莲1组与DDB治疗组、模型组大鼠相比较:芪莲1组肝功能明显好于同期的其他各组,纤维化程度轻。芪莲1组与芪莲2组、芪莲3组大鼠相比较:芪莲1组肝功能好于其他2组。芪莲2组以上观测指标均好于芪莲3组。结论具扶正祛毒功用的芪莲纤化汤对CCl4模型大鼠纤维化进展有明显延缓作用,并且干预得越早越好。
Objective To investigate the optimal timing of administration of Fuzheng Qudu drug to retard the progress of hepatic fibrosis. Methods Rat model of hepatic fibrosis was established by subcutaneous injection of CCl4 for 12 weeks. The Chinese herb group was treated with Qilianxianhua Decoction. The rats were divided into four groups according to the time of administration: the first week (Qilian group 1), the fourth week (Qilian group 2), 7th week (Qilian group 3) and western medicine group (DDB group) were treated with bifendate dropping pills on the first day of the first week. Different groups The liver biochemical indexes and liver histopathology of serum ALT, AST and ALB in different periods and different periods were also analyzed. Results After injection of CCl4, the general condition of rats in the model group gradually became worse, the weight of the rats in the model group decreased than that in the normal group, and the ascites appeared in the later period. The aminotransferase increased significantly from the 6th week, and the serum albumin decreased significantly at the 9th week. Obvious fiber spacing, hepatic lobules were destroyed, disorders, 9 weeks wide distribution of intrahepatic false lobules. Qilian 1 group and DDB treatment group, model group rats: Qilian group 1 liver function was significantly better than the other groups in the same period, the degree of fibrosis is light. Qilian 1 group and Qilian 2 group, Qilian 3 groups of rats compared: Qilian group 1 liver function better than the other two groups. Qilian 2 or more observations were better than Qilian 3 groups. Conclusion Qilianxianghua Decoction, which has the function of righting and removing disability, has a significant retardation on the progression of fibrosis in CCl4 model rats, and the sooner the better the intervention is.
