目的利用体外培养的大鼠乳鼠心肌细胞,探讨葛根素(puerarin,Pue)对异丙肾上腺素(isoproterenol,ISO)所致心肌细胞肥大和心肌细胞凋亡的影响。方法新生大鼠心肌细胞培养,除对照组外,分别给予ISO(10-5mol/L)、Pue(10-6mol/L)、ISO(10-5mol/L)+Pue(10-6mol/L)、ISO(10-5mol/L)+普萘洛尔(propranolol,Pro,2×10-6mol/L),检测培养心肌细胞蛋白质含量、细胞体积、蛋白质的合成及细胞凋亡率。结果和对照组相比,ISO可使心肌细胞总蛋白含量、体积和凋亡率明显增加(P<0.01);Pue可以抑制由于ISO诱导所出现的上述情况,结果与Pro相近。结论 Pue能对抗ISO引起的心肌肥大及细胞凋亡,此效应可能是葛根素抑制心肌肥厚发生的保护机制之一。
Objective To investigate the effect of Puerarin on the cardiomyocyte hypertrophy and cardiomyocyte apoptosis induced by isoproterenol (ISO) in rat neonatal rat cardiomyocytes. Methods The cultured neonatal rat cardiomyocytes were cultured in vitro with 10-5mol / L ISO, 10-6mol / L ISO, 10-6mol / L ISO and 10-6mol / , ISO (10-5mol / L) and propranolol (Pro, 2 × 10-6mol / L) were used to detect the protein content, cell volume, protein synthesis and apoptosis rate of cultured myocardial cells. Results Compared with the control group, ISO significantly increased the total protein content, the volume and the apoptosis rate of cardiomyocytes (P <0.01). Puerarin inhibited the above-mentioned events induced by ISO induction and the results were similar to Pro. Conclusion Puerarin can antagonize the myocardial hypertrophy and apoptosis induced by ISO. This effect may be one of the protective mechanisms by which puerarin inhibits cardiac hypertrophy.
