目的为了萃取和分析山豆根多糖组成及其单糖组分的构成,建立高效液相色谱法(HPLC)测定山豆根多糖中的单糖组分的分析方法。方法利用微波辅助萃取结合三氟乙酸水解分别获得山豆根粗多糖及其水解产物单糖;采用1—苯基—3—甲基—5—吡唑啉酮(PMP)衍生化—HPLC法测定。结果所建立的PMP衍生化—HPLC法可准确地测定山豆根多糖中葡萄糖、甘露糖、鼠李糖、葡萄糖醛酸、氨基半乳糖、半乳糖和阿拉伯糖7种单糖组分,其线性范围、检测限、加标回收率和相对标准偏差分别为1.00~500μg/mL、0.101~0.213μg/mL、98.3%~102.3%和0.39%~1.3%之间。结果表明该多糖为这7种单糖组成的杂多糖,其摩尔比组成为:葡萄糖∶阿拉伯糖∶半乳糖∶甘露糖∶葡萄糖醛酸∶鼠李糖∶氨基半乳糖=60.0∶17.3∶6.69∶5.27∶4.25∶4.04∶2.45。
OBJECTIVE To extract and analyze the composition of polysaccharides from Radix et Rhizoma radish and the composition of its monosaccharide components, an analytical method for the determination of monosaccharide components in Radix et Rhizoma with Radix et Rhizoma with Radix et Rhizoma rubrum was established by high performance liquid chromatography (HPLC). Methods The microwave-assisted extraction and trifluoroacetic acid hydrolysis were used to obtain the polysaccharides of R. sojae and their monosaccharides, respectively. The derivatization-HPLC method of 1-phenyl-3-methyl-5-pyrazolone (PMP) . Results The PMP-derivatization-HPLC method was established to accurately determine the content of seven monosaccharides, such as glucose, mannose, rhamnose, glucuronic acid, galactosamine, galactose and arabinose, The range, limit of detection, spike recoveries and relative standard deviations were 1.00 ~ 500μg / mL, 0.101 ~ 0.213μg / mL, 98.3% ~ 102.3% and 0.39% ~ 1.3%, respectively. The results showed that the polysaccharides were heteropolysaccharides composed of these seven kinds of monosaccharides with molar ratio of glucose: arabinose: galactose: mannose: glucuronic acid: rhamnose: galactosamine = 60.0:17.3:6.69: 5.27: 4.25: 4.04: 2.45.
