目的:确定感冒清热颗粒的最优制备工艺(每袋装3 g)。方法采用CO2-超临界流体法提取挥发油,以提油率为指标,优化萃取工艺,正交实验设计确定挥发油β-环糊精(β-CD)包合的最佳工艺,以葛根素转移率为指标,优选提取工艺,采用干法制粒方法,制粒。结果挥发油提取条件最佳工艺为:萃取压力20 MPa、萃取温度35℃、CO2流量为30 kg/h的条件下萃取120 min;环糊精最佳包合工艺为:油与β-CD比例为1:8,恒温40℃,搅拌包合1.5 h;煎煮最佳工艺为:加8倍水,煎煮2次,每次1.5 h。结论感冒清热颗粒的制备工艺稳定、可行。“,”Objective To detect optimal preparation of Ganmao Qingre Granule (3 g per bag). Methods Volatile oil was extracted by CO2- supercritical fluid.The extraction technology was optimized targeting on oil extraction rate.The best process of volatile oil β-cyclodextrin (β-CD) inclusion was determined by orthogonal experimental design.Puerarin transfer rate as an indicator,extraction process was optimized.The granulation was processed by dry granulating. Results The optimal process on volatile oil extracting was 20 MPa in extraction pressure,35℃ in extraction temperature,and 120-minnute extraction under the condition of 30 kg/h CO2 flow rate.The best inclusion process of cyclodextrin was that the ratio of oil andβ-CD being 1:8,40℃ in constant temperature,and stirring inclusion for 1.5 h.The optimal decoction process included 8 times of water,twice in decoction,and 1.5 h each time. Conclusion Preparation technics of Ganmao Qingre Granule is stable and feasible.
