目的 :研究钩藤碱 (Rhy)对大鼠脑缺血 -再灌注皮层和海马一氧化氮合酶 (NOS)变化的作用。方法 :阻断大鼠双侧颈总动脉 1 5min和再灌注 2 4h建立脑缺血 -再灌注损伤模型 ,用NADPH -d组化技术检测脑NOS阳性细胞数目的变化和Rhy的影响。结果 :脑缺血 -再灌注 2 4h后 ,皮层和海马NOS阳性细胞数目显著增多 ,Rhy( 1 2 .5和2 5mg/kg)缺血前 3 0min腹腔注射 ,可显著抑制此增多 ,与NOS拮抗剂L -NAME 1 0mg/kg的作用相似。结论 :Rhy能降低脑内NOS的活性 ,对脑缺血可能产生保护作用
Objective: To investigate the effect of Rhy on the changes of nitric oxide synthase (NOS) in the cortex and hippocampus after cerebral ischemia-reperfusion in rats. Methods: The model of cerebral ischemia-reperfusion injury was established by blocking bilateral common carotid arteries for 15 min and reperfusion for 24 h. The changes of NOS positive cells and Rhy were detected by NADPH-d staining. Results: The numbers of NOS positive cells in cortex and hippocampus increased significantly after cerebral ischemia-reperfusion for 24 h. Rhy (12.5 and 25 mg / kg) intraperitoneally injected 30 min before ischemia could significantly inhibit the increase of NOS positive cells, The effect of the antagonist L -NAME 10 mg / kg was similar. Conclusion: Rhy can reduce the activity of NOS in brain and may have a protective effect on cerebral ischemia
