目的:考察辐射剂量对石杉碱甲药剂学性质的影响。方法:石杉碱甲微球采用~(60)Co照射γ射线灭菌,采用高效液相色谱测定石杉碱甲微球的含量,通过透析释药法测定微球的体外释放度。采用扫描电镜观察微球的外观形态。结果:甘露醇按照其和微球0.5:1的比例加入,可以防止微球黏连,10,15,20 kGy的辐射剂量对微球的外观、含量及释放度无明显影响。结论:石杉碱甲微球采用辐射灭菌的方法可行。
Objective: To investigate the effect of radiation dose on the pharmacological properties of Huperzine A. METHODS: Huperzine A microspheres were sterilized by ~(60)Co irradiation with gamma rays. The content of Huperzine A microspheres was determined by high performance liquid chromatography. The in vitro release of the microspheres was determined by dialysis release method. Scanning electron microscopy was used to observe the morphology of the microspheres. RESULTS: Mannitol was added in a ratio of 0.5:1 to the microspheres to prevent the microspheres from adhering. Radiation doses of 10, 15, and 20 kGy had no significant effect on the appearance, content, and release of the microspheres. Conclusion: The method of radiation sterilization of huperzine A microspheres is feasible.
