目的:探讨槐定碱对内毒素致急性感染小鼠的免疫调节及氧化损伤保护作用。方法:采用随机数字法,将昆明小鼠60只分为6组:正常组、模型组、预防给药组、槐定碱低、中、高剂量组,每组10只。预防给药组连续6 d ip槐定碱5 mg.kg-1,其余各组ip 0.5 mL/只生理盐水,均末次给药后1 h除A组外其他各组一次性ip内毒素9 mg.kg-1建立小鼠急性感染模型。造模后2 h,槐定碱低、中、高剂量组分别ip槐定碱2.5,5,9 mg.kg-1,观察小鼠行为学变化;进行肺含水量的测定;取血检测白介素6(IL-6)水平和超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)的变化;HE染色观察肺组织细胞的形态改变。结果:给槐定碱各组小鼠的急性感染程度较模型组有明显改善,血清IL-6,MDA,NO含量较模型组降低(P<0.05);SOD含量较模型组升高(P<0.05);中剂量组效果优于其他各组(P<0.05)。HE染色提示,各给药组炎症程度均较模型组有不同程度的改善。结论:槐定碱可不同程度的减轻内毒素肺损伤小鼠的病理损害,能有效地增强模型鼠免疫调节能力及抗氧化损伤能力。
Objective: To investigate the immunomodulatory effect of sophoridine on endotoxin-induced acute infection in mice and the protective effect of oxidative damage. Methods: Sixty Kunming mice were randomly divided into six groups: normal group, model group, prophylaxis group and sophoridine low, medium and high dose groups, 10 in each group. The rats in prevention group were given ip sophoridine 5 mg.kg-1 for 6 days and ip 0.5 mL / normal saline in other groups, except for group A, kg-1 to establish a mouse acute infection model. 2 h after modeling, sophoridine low, medium and high dose groups were sophoridine 2.5, 5, 9 mg.kg-1, observed behavioral changes in mice; determination of lung moisture content; detection of interleukin 6, IL-6, SOD, MDA and NO were observed. The morphological changes of lung cells were observed by HE staining. Results: The acute infection in sophoridine group was significantly improved compared with the model group, the serum IL-6, MDA and NO levels were lower than the model group (P <0.05); the SOD content was higher than the model group (P < 0.05). The effect of middle dose group was better than other groups (P <0.05). HE staining suggested that the degree of inflammation in each administration group was improved to some extent compared with the model group. Conclusion: Sophoridine can alleviate the pathological damage of LPS-induced lung injury in mice to varying degrees, and can effectively enhance the immunomodulatory capacity and anti-oxidative damage ability in model mice.
