目的:观察人重组IL-8 在体内对新西兰家兔外周血白细胞的动员作用及其时间动力学。方法:按10μg/kg 的剂量在兔静脉内注射rhIL-8 ,收集不同时间的外周血,显微镜观察各种白细胞数量的变化。结果:IL-8 可升高外周血的白细胞数量,尤其是中性粒细胞的数量,从2.66 ×109/L升高至5 .99×109/L。IL- 8 动员的中性粒细胞以幼稚中性粒细胞(带状核)为主。IL-8 对中性粒细胞动员作用迅速,60min 时达到高峰,但持续时间短,180min 时已接近正常水平。结论:IL- 8 可显著升高外周血中性粒细胞数,而且是一种快速、安全的动员剂。
OBJECTIVE: To observe the mobilization and time kinetics of human recombinant interleukin-8 in peripheral blood leukocytes in New Zealand rabbits in vivo. Methods: The rhIL-8 was injected intravenously into rabbits at the dose of 10μg / kg. Peripheral blood was collected at different times, and the changes of the number of leukocytes were observed under a microscope. Results: IL-8 increased the number of white blood cells in peripheral blood, especially the number of neutrophils, from 2.66 × 109 / L to 5. 99 × 109 / L. The neutrophils mobilized by IL-8 are predominantly naive neutrophils (ribbon nuclei). IL-8 on the neutrophil mobilization rapid, peaked at 60min, but the duration is short, 180min was close to normal levels. Conclusion: IL-8 can significantly increase the number of peripheral blood neutrophils, but also a fast and safe mobilization agent.
