目的观察脑缺血再灌流后海马区cGMP的变化与细胞凋亡的分布 ,探讨两者的关系。方法钳夹沙士鼠的双侧颈总动脉制造脑缺血模型 ,应用免疫荧光组织化学法及TUNEL染色分别观察前脑缺血5min再灌流1d、2d、3d、4d后海马区cGMP的变化与细胞凋亡的分布。结果脑缺血再灌流后cGMP阳性细胞主要分布在放射层、腔隙分子层 ,且多数cGMP阳性细胞是星型胶质细胞 ,CA1 区锥体细胞未见cGMP阳性细胞。细胞凋亡全部分布在CA1 区锥体细胞。结论cGMP阳性细胞可能比cGMP阴性细胞缺血耐受。
Objective To observe the changes of cGMP and the distribution of apoptosis in hippocampus after cerebral ischemia and reperfusion, and to explore the relationship between them. Methods The bilateral common carotid arteries were sandwiched to produce cerebral ischemia model. The changes of cGMP in hippocampus were observed by immunofluorescence histochemistry and TUNEL staining at 1, 2, 3d and 4d after reperfusion Distribution of apoptosis. Results The cGMP positive cells mainly distributed in the radioactive layer and lacunar layer after cerebral ischemia and reperfusion, and most cGMP positive cells were astrocytes. No cGMP positive cells were found in CA1 pyramidal cells. Apoptosis are all distributed in CA1 pyramidal cells. Conclusion cGMP positive cells may be more resistant to ischemia than cGMP negative cells.
