①目的 探讨高压电子显微镜观察厚切片标本的技术。②方法 采用锌 碘 锇酸染色方法及AcPase(酸性磷酸酶 )细胞化学技术 ,制作高压电子显微镜观察的厚切片标本。③结果 高压电镜下显示 ,锌 碘 锇酸染色方法制备的标本胃底腺壁细胞的微管泡系统呈无数纤维状结构 ;AcPase结合三维电子显微镜观察显示 ,线性线粒体在胞质中常呈分支状并互相吻合成网。④结论 通过高压电子显微镜观察厚切片标本可以观察到细胞器的三维空间结构
①Objective To investigate the technique of high-pressure electron microscopy in the observation of thick sectioned specimens. Methods Zinc-iodine osmic acid staining method and AcPase (acid phosphatase) cytochemistry were used to make the thick slice specimens observed by high-voltage electron microscopy. Results The results of high pressure electron microscopy showed that the microtubule system of gastric fundus glandular parietal cells prepared by zinc-iodine-osmic acid staining showed numerous fibrous structures. AcPase combined with three-dimensional electron microscopy showed that the mitochondria linearly branched in the cytoplasm Anastomosis with each other into the network. ④ Conclusion Observed by high-pressure electron microscopy thick section of the specimen can be observed organelles three-dimensional spatial structure
