研究TNF介导的BCMEC释放PDGF及药物对BCMSMC增殖的保护作用.方法:体外培养BCMEC和BCMSMC,用结晶紫染色法观察TNF引起的BCMEC释放PDGF继而促进BCMSMC增殖作用.结果:TNF不能促进无血清培养的BCMSMC增殖,但TNF(5-20μg·L-1)剂量依赖地促进BCMEC释放PDGF.TNF(20μg·L-1)促进BCMSMC增殖的百分率为34±4%.药物Imperatorin,isoimperatorin和PMDP不影响TNF引起BCMEC释放PDGF,但能剂量依赖地(1-100μmol·L-1)抑制PDGF促BCMSMC增殖的作用.结论:TNF促进BCMEC释放PDGF继而引起BCMSMC的增殖.药物能抑制PDGF引起的BCMSMC增殖.
To study the protective effect of TNF-mediated release of PDGF by BCMEC and drug on the proliferation of BCMSMC. Methods: BCMEC and BCMSMC were cultured in vitro. Crystal violet staining was used to observe the release of PDGF by BCMEC induced by TNF and then the proliferation of BCMSMC. Results: TNF could not promote the proliferation of BCMSMC in serum-free medium, but TNF (5-20μg · L-1) promoted the release of PDGF in BCMEC in a dose-dependent manner. The percentage of proliferation of BCMSMC promoted by TNF (20μg · L-1) was 34 ± 4%. Drugs Imperatorin, isoimperatorin and PMDP did not affect TNF release of PDGF by BCMEC, but dose-dependently (1-100μmol·L-1) inhibited the proliferation of PDGF-stimulated BCMSMC. CONCLUSION: TNF promotes the release of PDGF by BCMEC and subsequently induces the proliferation of BCMSMC. Drugs can inhibit PDGF-induced BCMSMC proliferation.
