背景:黏着斑蛋白与心脏的先天畸形发育和心肌细胞的分化有着密切的关系。目的:观察α-黏着斑蛋白在大鼠心脏的表达分布特征以及在心肌细胞发育过程中随增龄发生的变化规律。设计:随机对照,观察对比实验。单位:新乡医学院细胞生物学教研室;濮阳职业技术学院生物工程与农业经济系。材料:实验于2003-01/2003-06在新乡医学院形态学研究室完成。选取清洁级Wistar大鼠28只,按年龄分为幼年组、青年组、中年组、老年组,每组7只。方法:所有大鼠在麻醉状态下取大鼠心脏组织,冠状连续系列切片,片厚5μm。幼年、青年、中年和老年大鼠的心肌细胞α-黏着斑蛋白的表达应用免疫组化方法进行观测。在细胞表面、胞浆中及闰盘处出现棕黄色颗粒为阳性细胞。所有标本均做常规染色(苏木精-伊红染色)为结构对照。主要观察指标:各组大鼠心肌细胞α-黏着斑蛋白的表达。结果:28只大鼠均进入结果分析。①α-黏着斑蛋白在大鼠的心房、心室、乳头肌、室间隔等处的心肌细胞均有表达。②幼年时期α-黏着斑蛋白的免疫标记颗粒大部分集中在心肌细胞末端的闰盘处,少量分散地分布于细胞表面和胞浆中;青年、中年和老年大鼠α-黏着斑蛋白的免疫标记典型的分布在心肌细胞末端的闰盘处。结论:在大鼠心肌发育过程中,α-黏着斑蛋白的表达分布存在增龄变化;α-黏着斑蛋白的分布方式与闰盘的发育是一致的。
Background: Corticotropin has a close relationship with the development of congenital malformations and cardiomyocytes. OBJECTIVE: To observe the distribution and distribution of α-plaque protein in rat heart and the changes of α-plaque protein during cardiomyocyte development with age. Design: Randomized, observational comparison experiment. Unit: Department of Cell Biology, Xinxiang Medical College; Department of Bioengineering and Agricultural Economics, Puyang Vocational and Technical College. MATERIALS: The experiment was performed in the Morphology Laboratory of Xinxiang Medical College from January 2003 to June 2003. Totally 28 Wistar rats were selected and divided into seven groups according to their age: young group, young group, middle age group and old age group. Methods: All rats were anesthetized to take the rat heart tissue, coronary continuous series of slices, slice thickness 5μm. The expression of α-plaque protein in cardiomyocytes of young, middle-aged and aged rats was observed by immunohistochemistry. In the cell surface, cytoplasm and intercalated disc appears brown granules are positive cells. All specimens were routinely stained (hematoxylin-eosin staining) as a structural control. MAIN OUTCOME MEASURES: Expression of α-plaque protein in rat cardiac myocytes. Results: All 28 rats were involved in the result analysis. ①α-adhesin protein in rat atria, ventricles, papillary muscles, ventricular septal and other places of myocardial cells are expressed. ② The immunolabeling particles of α-plaque protein were mostly concentrated in the intercalated disc at the end of cardiomyocytes, distributed in a small amount in the cell surface and in the cytoplasm in juvenile period; the expression of α-plaque protein in young, middle-aged and aged rats Immunological markers are typically found at the intercalated disc at the end of cardiomyocytes. CONCLUSION: The expression of α-plaque protein changes with aging in the process of myocardial development in rats. The distribution of α-plaque protein is consistent with the development of intercalated disc.
