目的观察氟伐他汀对动脉粥样硬化(As)家兔模型血管平滑肌细胞(SMC)增殖及血小板源性生长因子-A(PDGF-A)表达的影响。方法应用免疫组化法观察SMC增殖细胞核抗原(PCNA)表达,应用免疫组化及RT-PCR检测PDGF-A蛋白及mRNA表达。结果氟伐他汀组与高脂饮食组相比As病变明显减轻(P<0.01),PCNA表达明显减弱(P<0.01),PDGF-A蛋白及mRNA表达明显减弱(P<0.01)。PDGF-A蛋白表达与PCNA表达呈正相关(P<0.05)。结论氟伐他汀抗动脉粥样硬化作用机制可能与抑制PDGF-A表达,进而抑制SMC增殖有关。
Objective To observe the effect of fluvastatin on the proliferation of vascular smooth muscle cells (SMC) and the expression of platelet-derived growth factor-A (PDGF-A) in atherosclerotic rabbit models. Methods The expression of proliferating cell nuclear antigen (PCNA) in SMC was observed by immunohistochemistry. The expression of PDGF-A protein and mRNA was detected by immunohistochemistry and RT-PCR. Results Compared with the high-fat diet group, the As lesion in fluvastatin group was significantly reduced (P <0.01) and the expression of PCNA was significantly decreased (P <0.01). The protein and mRNA expression of PDGF-A in fluvastatin group was significantly decreased (P <0.01). PDGF-A protein expression was positively correlated with PCNA expression (P <0.05). Conclusion The anti-atherosclerotic effect of fluvastatin may be related to the inhibition of PDGF-A expression and the inhibition of SMC proliferation.
