目的探讨TPO、GATA-1和NF-E2在特发性血小板减少性紫癜(ITP)患儿骨髓表达水平的改变及其意义。方法分别收集28例急性ITP(AITP)患儿、19例慢性ITP(CITP)患儿和21例对照组儿童的骨髓;用Percoll密度梯度及免疫磁珠法分离骨髓巨核细胞;采用ABC免疫细胞化学染色法检测巨核细胞GATA-1和NF-E2的表达水平;ELISA法检测骨髓TPO的含量。结果AITP与CITP组骨髓巨核细胞GATA-1和NF-E2的表达水平均明显高于对照组(P<0.05),且CITP组的表达明显高于AITP组(P<0.05);AITP与CITP组的TPO浓度高于对照组,CITP组明显高于AITP组(P<0.05),三组之间无显著差异(P>0.05)。结论TPO?GATA-1和NF-E2均参与了巨核细胞生成的调控及ITP的发病过程。检测三者的表达水平可能对ITP的早期分型有意义。
Objective To investigate the changes and significance of bone marrow expression of TPO, GATA-1 and NF-E2 in children with idiopathic thrombocytopenic purpura (ITP). Methods Bone marrow of 28 children with acute ITP (AITP), 19 children with chronic ITP (CITP) and 21 control children were collected respectively. The bone marrow megakaryocytes were isolated by Percoll density gradient and immunomagnetic beads method. ABC immunocytochemistry The expression of GATA-1 and NF-E2 in megakaryocytes was detected by staining, and the content of TPO in bone marrow was detected by ELISA. Results The expression levels of GATA-1 and NF-E2 in bone marrow megakaryocytes of AITP and CITP groups were significantly higher than that of the control group (P <0.05), and significantly higher in the CITP group than in the AITP group (P <0.05) The TPO concentration was higher in the CITP group than in the AITP group (P <0.05). There was no significant difference between the three groups (P> 0.05). Conclusion Both TPO GATA-1 and NF-E2 are involved in the regulation of megakaryocytopoiesis and the pathogenesis of ITP. Detecting the expression levels of the three may have implications for the early typing of ITP.
