目的研究山楂叶总黄酮对异丙肾上腺素致大鼠急性心肌缺血的保护作用及其作用机制。方法取健康SD大鼠,♂,48只,随机分为正常对照组、阳性对照组、模型组及山楂叶总黄酮组(50,100,200 mg.kg-1),采用异丙肾上腺素(给药剂量10 mg.kg-1,每日1次,连续2 d)造成大鼠急性心肌缺血模型,观察急性心肌缺血大鼠心电图的变化,测定血浆的乳酸脱氢酶(LDH)和肌酸激酶(CK)、心肌组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和脂质过氧化物丙二醛(MDA)的含量,并观察心肌缺血大鼠心肌形态学变化。结果与模型组相比,山楂叶总黄酮(50,100,200 mg.kg-1)均可有效对抗异丙肾上腺素所致急性大鼠心肌缺血心电图ST段的抬高,降低血浆LDH和CK活力(P<0.05或P<0.01),增加心肌组织CAT和SOD的活力(P<0.05或P<0.01),并减少MDA生成(P<0.05或P<0.01),减轻心肌病理变化。结论山楂叶总黄酮对异丙肾上腺素致大鼠急性心肌缺血具有一定的保护作用,其机制可能与其抗氧化作用有关。
Objective To study the protective effect of hawthorn leaves flavonoids on isoproterenol-induced acute myocardial ischemia in rats and its mechanism. Methods Forty-eight healthy SD rats were randomly divided into normal control group, positive control group, model group and hawthorn leaf flavonoids group (50, 100, 200 mg.kg-1) and isoproterenol mg.kg-1, once a day for 2 days) induced acute myocardial ischemia in rats. The changes of electrocardiogram in acute myocardial ischemia rats were observed. The level of plasma lactate dehydrogenase (LDH) and creatine kinase CK, myocardial SOD, CAT, and lipid peroxides malondialdehyde (MDA) in myocardial tissue were measured. The morphological changes of myocardium were observed. Results Compared with the model group, total flavonoids of hawthorn leaves (50, 100, 200 mg.kg-1) could effectively resist the elevation of ST segment in ischemic myocardium induced by isoprenaline and decrease the activities of plasma LDH and CK <0.05 or P <0.01), and increased the activities of CAT and SOD (P <0.05 or P <0.01), decreased the production of MDA (P <0.05 or P <0.01) and decreased the pathological changes of myocardium. Conclusion The hawthorn leaves flavonoids have a protective effect on isoproterenol-induced acute myocardial ischemia in rats, and its mechanism may be related to its antioxidative effect.
