整合素是一类跨膜受体,是由α和β两个亚单位组成的异二聚体,主要通过与细胞外基质(extracellular matrix,ECM)相互作用将外界信号传入细胞内,并且在细胞生存、迁移、分化等多种细胞生命活动中发挥关键作用。具有多向分化潜能的间充质干细胞(mesenchymal stem cells,MSCs)的分化依赖整合素参与的信号通路,涉及MAPK、Wnt、PI3K信号通路等,而整合素的表达与活化受多种因素影响,如纳米形态微环境、抗坏血酸、骨成型蛋白-2、纤连蛋白、钙粘素、流体切应力等。本文主要对整合素表达的影响因素、整合素促进MSCs分化及其机制方面进行综述。
Integrins, a class of transmembrane receptors, are heterodimers composed of two subunits, alpha and beta, which transduce external signals into the cell primarily through interaction with the extracellular matrix (ECM) Cell survival, migration, differentiation and other cell life activities play a key role. The differentiation of mesenchymal stem cells (MSCs) with multilineage differentiation depends on integrin-involved signaling pathways, including MAPK, Wnt and PI3K signaling pathways. The expression and activation of integrins are influenced by many factors, Such as nano-morphology microenvironment, ascorbic acid, bone morphogenetic protein-2, fibronectin, cadherin, fluid shear stress and the like. This article mainly summarizes the influencing factors of integrin expression, integrins promote MSCs differentiation and its mechanism.
