目的建立以高效液相色谱法测定口服塞克硝唑胶囊后人血液和唾液中塞克硝唑含量的方法。方法以奥硝唑为内标,乙腈沉淀蛋白后直接进样。色谱柱为Zorbax SB-C18,流动相为0.05mol·L-1磷酸二氢钾(pH=5)-乙腈(75∶25),检测波长为311nm,流速为0.8mL·min-1。结果血液和唾液中塞克硝唑分别在1~80,0.5~80mg·L-1l范围内线性关系良好,最低检测浓度分别为0.084和0.0167mg·L-1,方法回收率为98.34%~109.01%,日内RSD≤3.6%,日间RSD≤4.0%,血液和唾液中塞克硝唑的绝对回收率均>88.58%。结论此法能简便、灵敏、准确地测定牙周炎患者血液和唾液中的浓度,可用于临床和科学研究。
Objective To establish a HPLC method for the determination of secnidazole in human blood and saliva after oral administration of Secnidazole capsules. Methods Ornidazole as an internal standard, acetonitrile precipitation protein directly after injection. The column was Zorbax SB-C18 with a mobile phase of 0.05 mol·L-1 potassium dihydrogen phosphate (pH = 5) -acetonitrile (75:25) at a detection wavelength of 311 nm and a flow rate of 0.8 mL · min-1. Results Selegitidine in blood and saliva showed good linearity in the range of 1 ~ 80,0.5 ~ 80 mg · L-1l with the lowest detection concentrations of 0.084 and 0.0167 mg · L-1, respectively. The recoveries were 98.34% -109.01 %, Intraday RSD≤3.6%, daytime RSD≤4.0%, and the absolute recoveries of selecidazole in blood and saliva were both> 88.58%. Conclusion This method can be simple, sensitive and accurate determination of the concentration of blood and saliva in patients with periodontitis can be used for clinical and scientific research.
