目的:采用近红外光谱透射法快速测定栀子逆流提取过程中总苷含量。方法:以UV分析值作为参照,采用NIR透射法结合偏最小二乘法建立总苷含量的快速在线测定方法。采样方式为透射采集,采集区间4 000~10 000 cm-1,分辨率8.0 cm-1,扫描数64次,empty门衰减,增益为1,温度(25±2)℃,相对湿度45%~50%。结果:建立的栀子逆流提取液中总苷校正模型的相关系数、校正均方差分别为0.993,0.179。经外部验证,校正模型的预测均方差0.145,预测值和真实值间回归方程的相关系数达0.991。结论:NIR可作为一种准确、快速的在线检测方法用于中药提取过程中有效成分的含量检测。
Objective: To determine the content of total glycosides in Gardenia countercurrent extraction by near-infrared spectroscopy. Methods: The method of NIR transmission and partial least squares was used to establish the on-line determination method of total glycosides by UV analysis. The sampling method is transmission collection, the sampling interval is 4 000 ~ 10 000 cm-1, the resolution is 8.0 cm-1, the scanning number is 64 times, the empty door attenuation is 1, the temperature is 25 ± 2 ℃ and the relative humidity is 45% 50%. Results: The correlation coefficient and the corrected mean square error of the total glycoside calibration model established in Gardenia countercurrent extract were 0.993,0.179. According to the external validation, the predicted variance of the calibration model is 0.145, and the correlation coefficient between the predicted and the true values is 0.991. Conclusion: NIR can be used as an accurate and rapid on-line detection method for the determination of active ingredients in the process of Chinese traditional medicine extraction.
