为了探讨心钠素(ANF)对内皮素(ET)生物学效应的拮抗作用,我们进行了下述实验。在ET致小鼠死亡实验中,观察到大剂量ANF可显著延长小鼠存活时间;在ET致大鼠升压实验中,观察到ANF明显拮抗ET的升压作用;在大鼠离体胸主动脉环灌流实验中,观察到ANF对ET引起的血管收缩呈浓度依赖的舒张作用;在培养的血管平滑肌细胞中,观察到ANF显著抑制ET的促血管平滑肌细胞钙内流作用。结果表明:ANF对ET生物学效应具有部分拮抗作用,ANF在治疗与ET有关的疾病上,可能具有潜在临床意义。
In order to investigate the antagonism of atrial natriuretic peptide (ANF) on the biological effects of endothelin (ET), we performed the following experiment. In the ET-induced mouse death experiment, it was observed that high-dose ANF can significantly prolong the survival time of mice; in the ET-induced rat vasopressor test, it was observed that ANF significantly antagonized the ET-induced vasopressor effect; In arterial ring perfusion experiments, it was observed that ANF attenuated the vasoconstriction induced by ET in a concentration-dependent manner. In cultured vascular smooth muscle cells, it was observed that ANF significantly inhibited the calcium influx in ET-stimulated vascular smooth muscle cells. The results showed that: ANF has partial antagonism on the biological effects of ET, and ANF may have potential clinical significance in the treatment of ET-related diseases.
