目的观察“热敷药”对硬皮病小鼠模型Ⅰ型胶原、Ⅲ型胶原(COL-Ⅰ、COL-Ⅲ)含量的影响。方法 60只BALB/c小鼠按照随机原则分为空白组、模型组、西药组、小浓度组、中浓度组、大浓度组各10只,采用博莱霉素皮下注射形成硬皮病模型,造模完成后,根据分组分别给予10%、20%、40%“热敷药”药糊及肝素钠软膏外用,1次/d,连续1个月,空白组及模型组不用药。完成后观察各组皮肤组织病理变化,免疫组化检测皮损COL-Ⅰ、COL-Ⅲ蛋白含量。结果与模型组相比较,高、中浓度组硬皮病小鼠模型COL-Ⅰ、COL-Ⅲ蛋白含量明显降低,差异较显著(P<0.05);结论“热敷药”具有改善硬皮病小鼠模型皮肤硬化,降低COL-Ⅰ、COL-Ⅲ的作用。
Objective To observe the effect of “Fuji” on the content of type Ⅰ collagen and type Ⅲ collagen (COL-Ⅰ, COL-Ⅲ) in mouse models of scleroderma. Methods Sixty BALB / c mice were divided into blank group, model group, western medicine group, small concentration group, medium concentration group and high concentration group according to the principle of randomness. 10 rabbits were injected subcutaneously with bleomycin to form scleroderma model, After modeling, 10%, 20%, 40% “hot drug ” drug paste and heparin sodium ointment were given for external use according to grouping, once a day for 1 month, and blank group and model group were given no drug. After completion of the observation of the pathological changes of skin tissue, immunohistochemical detection of lesions COL-Ⅰ, COL-Ⅲ protein content. Results Compared with model group, the contents of COL-Ⅰ and COL-Ⅲ in scleroderma model mice were significantly decreased (P <0.05) Sick mouse model of skin sclerosis, reducing the role of COL-Ⅰ, COL-Ⅲ.
