目的:以急性心肌梗死大鼠模型为研究对象,通过观察不同时间点四逆汤对该模型大鼠血浆中肌酸激酶(CK)、肿瘤坏死因子(TNF-α)活性的作用,探讨四逆汤对大鼠急性缺血性心肌梗死模型的CK和TNF-α动态药效影响。方法:将Wistar大鼠随机分为3组:心肌缺血组、假手术组、四逆汤组。心肌缺血组:左冠状动脉前降支起始部下2mm处结扎;假手术组:穿线不结扎;四逆汤组:预先给大鼠灌服四逆汤4天,第4天给药30分钟后,进行急性缺血性心肌梗死手术。按时间点0、5、10、20、40、60、90、120、150、180、210、360分钟采血,分别检测CK和TNF-α含量。结果:心肌缺血组、四逆汤组与假手术组比较,心肌缺血组的CK(10、20、60、90、120、150、360分钟)、TNF-α(120、150分钟),四逆汤组的CK(10、20分钟)含量明显升高,TNF-α含量无明显差异;四逆汤组与心肌缺血组比较,四逆汤组可显著降低血浆中CK(60、90、120、150、180、210、360分钟)、TNF-α(5、20、40、60、90、120、360分钟)的含量。假手术组、心肌缺血组CK含量与时间变化均为正相关关系,四逆汤组TNF-α含量与时间变化为负相关关系。结论:实验表明四逆汤能够有效地减少心肌酶的释放,改善缺血心肌的损伤,对大鼠缺血心肌具有积极明显的保护作用。各组间动态相关分析提示,通过降低炎性因子TNF-α含量,可能是四逆汤抗心肌缺血的重要机制之一。
Objective: To investigate the effect of Sini Decoction at different time points on the activity of creatine kinase (CK) and tumor necrosis factor (TNF) in plasma of rats with acute myocardial infarction (AMI) Effect of Decoction on CK and TNF-α in Acute Ischemic Myocardial Infarction Model Rats. Methods: Wistar rats were randomly divided into three groups: myocardial ischemia group, sham operation group and Sini decoction group. The rats in the ischemia group were ligated at 2mm below the anterior descending branch of the left coronary artery. Sham-operation group was not ligated by threading. The Sini decoction group was fed with Sini decoction for 4 days and the fourth day for 30 minutes After the operation of acute ischemic myocardial infarction. Blood samples were taken at 0, 5, 10, 20, 40, 60, 90, 120, 150, 180, 210 and 360 minutes to detect the contents of CK and TNF- Results: Compared with sham-operated group, myocardial ischemia group had significantly lower levels of CK (10,20,60,90,120,150,360 minutes), TNF-α (120,150 minutes) Sini Decoction group CK (10,20 minutes) content was significantly increased, TNF-α content was no significant difference; Sini Decoction group and myocardial ischemia group, Sini Decoction group can significantly reduce the plasma CK (60,90 120, 150, 180, 210, 360 minutes), TNF-a (5,20,40,60,90,120,360 minutes). There was a positive correlation between CK content and time in sham-operation group and myocardial ischemia group, and there was a negative correlation between the content of TNF-α and the time in Sini Decoction group. Conclusion: Experiments show that Sini Decoction can effectively reduce the release of myocardial enzymes, improve myocardial ischemia, has a positive protective effect on ischemic myocardium in rats. Dynamic correlation analysis among the groups suggested that it may be one of the important mechanisms of anti-myocardial ischemia of Sini Decoction by reducing the content of TNF-α.
