目的 :为比较不同组织切片方法对免疫组织化学结果的影响 .方法 :用成年SD大鼠L5脊髓分别制成冰冻 (2 0 μm)和石蜡 (5 μm)切片 ,用兔抗BDNF多克隆抗体作ABC免疫组化染色 .结果 :冰冻切片组背角Ⅱ板层除有大量BDNF阳性神经膨体外 ,尚可见一些阳性神经元 .石蜡切片组则仅有大量阳性神经膨体 ,而几乎未见阳性神经元 .结论 :不同制片法对免疫组织化学结果有影响 ,即冰冻切片可能较石蜡切片具有更高的免疫反应敏感性 .本文中石蜡切片较低的BDNF免疫活性可能与其制作过程中有机溶剂 (酒精、二甲苯 )的使用和 /或包埋过程中加热处理所致的BDNF抗原丢失有关
