以刨花润楠种子为外植体,采用丛芽发生途径,初步建立其组培快繁技术体系。结果表明,GA_3浸泡种子可显著提高发芽率,缩短发芽时间。子叶节最易诱导不定芽,诱导芽数最多。刨花润楠丛芽再生能力较弱,培养过程中愈伤组织多,整体情况较差。较为适宜的增殖方式为外植体在MS+6-BA 3.0 mg·~(L-1)+IBA 0.01 mg·~(L-1)+AgNO_3 1.0 mg·~(L-1)上预培养10 d,再转入MS+6-BA 3.0 mg·~(L-1)+IBA 0.01 mg·~(L-1)中培养,增殖系数可达3.52。生根培养基为1/2MS+IBA 0.2 mg·~(L-1),生根率可达91%。
Using Machilus lucidum seeds as explants, using the way of cluster buds, the tissue culture and propagation technology system was initially established. The results showed that GA 3 soaking seeds could significantly increase the germination rate and shorten the germination time.
