目的:研究不同品种的淫羊藿中4种单体成分的含量差异,及体外促大鼠颅骨成骨细胞增殖的作用。方法:采用HPLC法,测定淫羊藿样品中淫羊藿苷和朝藿定A、B、C的含量;以SD乳鼠颅骨二次酶消化法培养成骨细胞,采用MTT法,测定4种单体,模拟不同淫羊藿含量配比的单体组合物对成骨细胞的增殖作用。结果:8个药材中湖北朝鲜淫羊藿含淫羊藿苷和朝藿定A最高;在1.0-100.0μg/m L浓度范围内,淫羊藿苷显著促进成骨细胞的增殖,作用呈浓度依赖性,朝藿定A显著促进成骨细胞的增殖,作用与浓度负相关,朝藿定B能促进成骨细胞增殖,朝藿定C对成骨细胞的增殖影响不大;各药材模拟组在0.1-10.0mg/m L生药浓度范围内,淫羊藿苷低浓度时能显著促进成骨细胞增殖,高浓度时增殖作用有所抑制,朝藿定B对成骨细胞增殖作用随浓度增大而增强,朝藿定C无明显增殖活性。结论:淫羊藿苷和朝藿定A、B单体均具有促进成骨细胞增殖活性;淫羊藿促成骨细胞增殖作用与4种成分的含量以及配比密切相关。
OBJECTIVE: To study the content differences of four kinds of monomers in different varieties of Epimedium, and to promote the proliferation of rat calvarial osteoblasts in vitro. Methods: HPLC method was used to determine the contents of icariin and epimedin A, B and C in epimedium samples. The osteoblasts were cultured with the second enzyme digestion method of SD suckling mice. MTT assay was used to determine the contents of icariin, Monomer to simulate different Epimedium content of the monomer composition of the proliferation of osteoblasts. Results: The Epimedium koreanum contained the highest concentrations of Epimedium and Epimedin A in eight medicinal materials. Icariin significantly promoted the proliferation of osteoblasts in the concentration range of 1.0-100.0μg / mL with a concentration of Dependency, Epimedin A significantly promote the proliferation of osteoblasts, the role of concentration and negatively related to epimedin B can promote osteoblast proliferation, Epimedin C on osteoblast proliferation was not significant; the medicinal simulation group In the concentration range of 0.1-10.0mg / m L crude drug, Icariin could significantly promote the proliferation of osteoblasts at low concentration, inhibited the proliferation at high concentration, and the effect of Epimedin B on the proliferation of osteoblasts increased with increasing concentration Large and enhance, Epimedin C no significant proliferative activity. CONCLUSION: Icariin and Epimedin A and B can promote the proliferation of osteoblasts. Epimedium promoted the proliferation of osteoblasts with the content and ratio of four components.
