本文将人胎盘用胃蛋白酶消化、氯化钠分级沉淀及DE-52、CM-52层析提取、纯化N M胶原(ⅣC)。提取物紫外最大吸收峰在230nm,富含羟脯氨酸、羟赖氨酸,两个α链分子量分别为95000和70000,与Sigma产品特性相似。免疫新西兰兔产生的抗血清效价为1:512(间接血凝法),与其它间质成份无或弱交叉反应。建立的放射免疫分析方法,灵敏度为8μg/L,批内及批间C.V.分别为7.3%和11.2%,测得正常献血员血清ⅣC浓度为49.77±15.00μg/L,慢性肝炎患者显著升高,并与PANASSAY ⅣC EIA法检测结果相关显著,提示本方法灵敏可靠,有推广应用价值。
In this paper, human placenta digestion with pepsin, sodium chloride fractionation and DE-52, CM-52 chromatography extraction, purification of N M collagen (Ⅳ C). The maximum ultraviolet absorption peak of the extract was at 230 nm, which was rich in hydroxyproline and hydroxylysine. The molecular weights of the two α-chains were 95000 and 70000, respectively, which were similar to those of Sigma products. The antiserum titer produced by immunized New Zealand rabbits was 1: 512 (indirect hemagglutination) with no or weak cross-reactivity with other interstitial components. The established method of radioimmunoassay showed that the sensitivity was 8μg / L, the intra-and inter-assay CV was 7.3% and 11.2%, respectively. The serum IVC concentration of normal blood donors was 49.77 ± 15.00μg / L, And with PANASSAY Ⅳ C EIA test results were significant, suggesting that the method is sensitive and reliable, and has the value of popularization and application.
