【目的】在全基因组水平上鉴定亚洲棉(Gossypium arboretum L.)AGO蛋白家族成员,分析其进化关系,为亚洲棉遗传改良提供基因资源。【方法】用AGO蛋白保守域在亚洲棉基因组数据库中进行同源比对,并对AGO蛋白家族的进化关系、基因结构、结构域、染色体定位、三级结构及表达模式等进行生物信息学分析。【结果】从亚洲棉蛋白数据库中共鉴定出11个AGO蛋白家族成员,其编码区长度为2 949~9 807bp,定位在6条染色体上,其中第9号染色体上分布有4个基因;11个AGO家族成员分为5个亚族,亚族内各成员间具有很高的保守性;AGO家族成员均具有典型的α-螺旋跨膜结构,属于碱性蛋白;从表达模式看,AGO家族成员在纤维发育不同时期和不同组织中的表达量存在较大差异,其中5个基因在棉纤维次生壁后期高表达,在其他时期表达水平较低,存在组织特异性。【结论】成功鉴定了亚洲棉AGO蛋白家族,明确了其基本结构的进化关系及基因表达特点。
【Objective】 The aim of the present study was to identify AGO protein family members of Gossypium arboretum L. at genome-wide level and to analyze their evolutionary relationships to provide genetic resources for genetic improvement in cotton. 【Method】 The homology alignment of the AGO protein conserved region in the genome database of Asian cotton was carried out and the bioinformatics analysis was carried out on the evolutionary relationship, gene structure, domain, chromosomal location, tertiary structure and expression pattern of AGO protein family . 【Result】 A total of 11 AGO protein family members were identified from the Asiatic cotton protein database. The length of the coding region was 2949-9807 bp, located on 6 chromosomes. There were 4 genes distributed on chromosome 9 and 11 AGO family members are divided into five sub-families, among the members of the sub-family is highly conserved; AGO family members have a typical α-helical membrane structure, belonging to basic protein; from the expression pattern, AGO family members There were significant differences in the expression of different genes in different stages of fiber development. Among them, 5 genes were highly expressed in the secondary wall of cotton fiber and low in other stages, and there was tissue specificity. 【Conclusion】 The AGO protein family of Asiatic cotton was successfully identified and the evolutionary relationship and gene expression characteristics of its basic structure were clarified.
