[目的]探讨细丝蛋白A(filamin A,FLNa)在肺鳞癌组织中的表达及其临床意义。[方法]采用Western Blot方法和逆转录聚合酶链反应(RT-PCR),分别检测43例肺鳞癌组织及距肺鳞癌组织边缘5cm以上的镜下未见癌组织浸润的正常组织中FLNa蛋白、mRNA的表达情况。[结果]FLNa蛋白在肺腺鳞组织及正常肺组织中的相对表达量分别为0.381±0.032、0.673±0.072,两组差异有统计学意义(P<0.05)。FLNa mRNA在肺腺鳞组织及正常肺组织中的相对表达量分别为0.356±0.052、0.573±0.075,两组差异有统计学意义(P<0.05)。FLNa蛋白、mRNA表达与肺鳞癌患者性别、年龄、肿瘤大小、肿瘤分化无关(P>0.05);而与淋巴结转移以及肿瘤TNM分期有关(P<0.05)。[结论]肺鳞癌中FLNa蛋白和mRNA的表达均降低,可能与肺鳞癌的发生、发展相关。
[Objective] To investigate the expression and clinical significance of filamin A (FLNa) in squamous cell lung cancer. [Methods] Western Blot and reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of FLNa in 43 squamous cell carcinoma of lung squamous cell carcinoma and 5 cm squamous cell carcinoma of lung squamous cell carcinoma Protein, mRNA expression. [Results] The relative expression levels of FLNa protein in lung adenosquamous and normal lung tissues were 0.381 ± 0.032 and 0.673 ± 0.072 respectively, with statistical significance (P <0.05). The relative expression levels of FLNa mRNA in lung adenosquamous and normal lung tissues were 0.356 ± 0.052 and 0.573 ± 0.075 respectively, with statistical significance (P <0.05). The expression of FLNa protein and mRNA was not associated with gender, age, tumor size, tumor differentiation (P> 0.05), but correlated with lymph node metastasis and tumor TNM stage (P <0.05). [Conclusion] The expression of FLNa protein and mRNA in lung squamous cell carcinoma are both decreased, which may be related to the occurrence and development of squamous cell carcinoma of the lung.
