目的:观察续骨活血汤对家兔骨折愈合中血清骨钙素(BGP)、骨形态蛋白-2(BMP-2)表达和血液流变学的影响。方法:将108只家兔建立骨折模型,随机分为模型组、伤科接骨片组和续骨活血汤组,每组36只,各组均于造模术后第2天开始给药,2次/d,模型组予蒸馏水灌胃,伤科接骨片组予伤科接骨片200 mg/kg以混匀灌胃,续骨活血汤组予续骨活血汤浓缩口服液对人等效剂量的 2 倍灌胃。术后2周时随机抽取各组6只家兔,用自动血黏度测试仪检测血液流变学指标。于术后1、2、3、4、6周时随机抽取各组6只家兔,采用放射免疫法测定血清中骨钙素含量,再取材,进行免疫组织化学染色,观察骨折愈合不同时间点外骨膜、内骨膜和骨髓腔骨BMP-2的表达情况。结果:血液流变学及BGP、BMP-2的表达,续骨活血汤组、伤科接骨片组大部优于模型组,P<0.01或P<0.05;续骨活血汤组与伤科接骨片组比较,血液流变学大部时间点优于伤科接骨片,P<0.01或P<0.05;BGP和BMP-2的表达,部分时间点高于伤科接骨片组,P<0.01或P<0.05。结论:续骨活血汤对家兔骨折愈合有较好促进作用。
OBJECTIVE: To observe the effects of “Decoction for Promoting Blood Circulation of Osseous and Exhaustion Decoction” on the expression of serum osteocalcin (BGP), bone morphogenetic protein-2 (BMP-2) and hemorheology in rabbits during fracture healing. Methods: A total of 108 rabbits were randomly divided into model group, Shangke Jiegu tablet group and XGJD group, with 36 rabbits in each group. The rats in each group were given the drug on the second day after model establishment. Times / d, the model group was given gavage with distilled water. The Shangkejiegu tablet group was given Shangkejiegu tablet (200mg / kg) 2 times gavage. At 2 weeks after operation, 6 rabbits in each group were randomly selected and their hemorheological indexes were measured by automatic blood viscosity tester. At 1, 2, 3, 4, and 6 weeks after operation, 6 rabbits in each group were randomly selected, and the serum osteocalcin levels were determined by radioimmunoassay. The specimens were harvested for immunohistochemical staining. Periosteum, endosteal and bone marrow cavity BMP-2 expression. Results: The hemorrheology, the expression of BGP and BMP-2 in XJGD group were better than model group, P <0.01 or P <0.05. P <0.01 or P <0.05; The expression of BGP and BMP-2 at some time points was higher than that in Shangke Jiegu Tablet group, P <0.01 or P <0.01 or P < P <0.05. CONCLUSION: CONGJDT can promote fracture healing in rabbits.
