目的 探讨转移抑制基因nm 2 3 H1在肝癌组织中的表达和癌细胞增殖及DNA含量间的关系。方法 应用免疫组织化学染色法检测 5 6例肝癌组织标本中nm 2 3 H1基因和增殖细胞核抗原 (PCNA)的表达 ,应用图像分析系统测定癌细胞DNA含量 ,分析与肝癌病理生物学行为之间的关系。结果 nm2 3 H 1表达阳性率无包膜组肝癌 (2 9.6% )比包膜完整 (64 .3 % )和包膜突破组肝癌 (66.7% )明显减低 (P <0 .0 5 )。DNA指数 (DI)与肝癌包膜情况、组织类型、组织分级也有显著相关性 (P <0 .0 5 )。nm 2 3 H1表达阴性的肝癌PCNA标记指数 (LI)高于nm 2 3 H1阳性者 (P <0 .0 5 ) ;PCNA标记指数高增殖组肝癌DI值 (2 .3 0± 0 .90 )较低增殖组肝癌DI值 (1.86± 0 .7)明显增高 (P <0 .0 5 )。结论 nm 2 3 H1表达与肝癌包膜形成具有一定关系 ,并与肝癌增殖活性相关。DNA含量测定结合PCNA免疫组织化学染色可较为准确的反映肝癌浸润侵袭特征和增殖活性
Objective To investigate the relationship between the expression of metastasis suppressor gene nm23H1 in hepatocellular carcinoma and the proliferation and DNA content of cancer cells. Methods Immunohistochemical staining was used to detect the expression of nm23H1 gene and proliferating cell nuclear antigen (PCNA) in 56 specimens of hepatocellular carcinoma. Image analysis system was used to determine the DNA content of the cancer cells, and the pathological biological behaviors of hepatocellular carcinoma were analyzed. relationship. Results The positive rate of nm2 3 H 1 expression in the non-enveloped group was significantly lower than that in the capsular group (64.6%) and in the capsular breakthrough group (66.7%) (P < 0.05). The DNA index (DI) was also significantly correlated with liver cancer capsule status, tissue type, and tissue grade (P < 0.05). The PCNA labeling index (LI) of liver cancer with negative expression of nm23H1 was higher than that of nm23H1 (P0.05); the DI value of liver cancer with high proliferation of PCNA index (2.30±0.90). The DI value of the lower proliferation group (1.86 ± 0.7) was significantly higher (P <0.05). Conclusion The expression of nm23H1 has a relationship with the formation of hepatocellular carcinoma capsule, and it is related to the proliferation activity of hepatocellular carcinoma. The determination of DNA content combined with PCNA immunohistochemical staining can accurately reflect the characteristics of invasion, invasion and proliferation of liver cancer.
