目的建立菊花中3,5-O-双咖啡酰基奎宁酸、木犀草苷、绿原酸3种成分定量测定的方法,并对4种菊花3种成分的量进行比较。方法采用高效液相色谱法在同一色谱条件(色谱柱为Shimadzu VP-ODS 250 mm×4.6mm,5μm;流动相:乙腈-0.1%磷酸梯度洗脱;体积流量:1.0 mL/min;柱温:30℃)下对菊花中3种成分同时进行定量测定。结果3,5-O-双咖啡酰基奎宁酸、木犀草苷、绿原酸分别在0.039 1~0.781 0μg(r=1.000 0),0.013 2~0.396 0μg(r=1.000 0),0.017 0~0.510 0μg(r=0.999 3)线性关系良好。3者的平均回收率分别为97.7%(RSD=2.7%),101.7%(RSD=1.5%),98.0%(RSD=1.2%)。结论所建立的方法可在同一色谱条件下检测出3个成分,4种菊花的3种成分量有不同程度的差别。
OBJECTIVE To establish a method for the quantitative determination of 3,5-O-bis-caffeoylquinic acid, luteolin, and chlorogenic acid in chrysanthemum and to compare the contents of three components in four chrysanthemums. Methods High performance liquid chromatography (HPLC) was used under the same chromatographic conditions (column: Shimadzu VP-ODS 250 mm × 4.6 mm, 5 μm; gradient elution with acetonitrile-0.1% phosphoric acid; volume flow rate: 1.0 mL / 30 ℃) chrysanthemum in the three components at the same time quantitative determination. Results The results showed that the concentrations of 3,5-O-bis-caffeoylquinic acid, luteolin and chlorogenic acid were in the ranges of 0.039 1-0.781 μg (r = 1.000 0), 0.013 2-0.396 μg (r = 1.000 0) 0.510 0μg (r = 0.999 3) Good linearity. The average recoveries of the three were 97.7% (RSD = 2.7%), 101.7% (RSD = 1.5%) and 98.0% (RSD = 1.2%), respectively. Conclusion The established method can detect three components under the same chromatographic conditions, and the three components of four kinds of chrysanthemum have different degrees of difference.
