目的 建立莪术挥发油中吉马酮在大鼠血浆中的高效液相色谱测定方法。方法 血浆样品经乙腈沉淀蛋白处理后 ,取上清液在 2 4 4nm波长下采用高效液相色谱法进行分析。结果 吉马酮在血中的检测限为 10 0ng·mL- 1 ,线性范围为0 10 0 4 - 15 0 6 μg·mL- 1 。日内和日间的RSD分别为 1 87% - 4 2 9%和 1 2 9% - 5 15 %。吉马酮的提取回收率为 95 %以上。血浆中的内源性物质对测定没有影响。结论 该法操作简便、快速 ,适用于吉马酮在体内的药代动力学研究
Objective To establish a method for the determination of germacrone in essential oil of Rhizoma Curcumae by HPLC with high performance liquid chromatography (HPLC). Methods After the plasma samples were treated with acetonitrile precipitation protein, the supernatant was analyzed by high performance liquid chromatography at a wavelength of 244nm. Results The detection limit of gemcitabine in blood was 10 ng · mL-1 with a linear range of 0 10 0 4 -15 0 6 μg · mL-1. The intra-day and inter-day RSDs were 1 87% - 42 9% and 1 2 9% - 5 15% respectively. The recovery of gemcitabine is more than 95%. Endogenous substances in plasma have no effect on the assay. Conclusion The method is simple, rapid and suitable for the study of pharmacokinetics of germacrone in vivo
