目的研究甘草甜素对人乳腺癌细胞MDA-MB-231生长的影响。方法将对数生长期人乳腺癌细胞随机分为5组:对照组仅加等量培养液,实验A组、B组、C组及D组分别给予浓度分别为0,50,100,200μmol·L~(-1)的甘草甜素细胞培养液;用噻唑蓝(MTT)法检测5组人乳腺癌细胞MDA-MB-231增殖、凋亡情况,并采用半定量逆转录-聚合酶联反应(RT-PCR)检测细胞中Fas、survivin、Bcl-2、Bax和caspase-3基因水平。结果随着甘草甜素浓度的增加和作用时间的延长,对人乳腺癌细胞MDA-MB-231增殖抑制作用显著增强,其中48 h后对照组OD值为(0.61±0.21),实验A、B、C、D组分别为(0.61±0.11),(0.56±0.15),(0.48±0.17),(0.45±0.14),实验A、B、C、D组的抑制率分别为8.17%,10.01%,24.41%,31.82%,各组比较差异有统计学意义(P<0.05);Fas、survivin、Bax和caspase-3基因表达因甘草甜素浓度增加而降低,而Bcl-2表达则因甘草甜素浓度增加而上升。实验D组的Fas、survivin、Bax和caspase-3基因表达下降程度明显高于其他组(P<0.05),Bcl-2基因表达水平增加程度也明显高于其他组(P<0.05)。结论甘草甜素对人乳腺癌细胞MDA-MB-231细胞增殖抑制作用强,具有明显的量效和时效关系。
Objective To study the effect of glycyrrhizin on the growth of human breast cancer cell line MDA-MB-231. Methods The human breast cancer cells in logarithmic growth phase were randomly divided into 5 groups: the control group was given only equal amount of culture solution, and the concentrations in group A, group B, group C and group D were 0, 50, 100, 200μmol·L ~ -1). The proliferation and apoptosis of 5 human breast cancer cell lines MDA-MB-231 were detected by MTT assay. The proliferation and apoptosis of MDA-MB-231 cells were detected by semi-quantitative reverse transcriptase-polymerase chain reaction (RT- PCR) was used to detect the expression of Fas, survivin, Bcl-2, Bax and caspase-3 in the cells. Results With the increase of glycyrrhizin concentration and prolongation of action time, the proliferation of human breast cancer cells MDA-MB-231 was significantly enhanced. The OD value of control group after 48 h was (0.61 ± 0.21) (0.61 ± 0.11), (0.56 ± 0.15), (0.48 ± 0.17) and (0.45 ± 0.14) in group C and D, respectively. The inhibition rates in groups A, B, C and D were 8.17% and 10.01% , 24.41% and 31.82%, respectively (P <0.05). The expressions of Fas, survivin, Bax and caspase-3 genes were decreased in response to the increase of glycyrrhizin concentration, while the expression of Bcl- Elemental concentration increased and increased. The expression of Fas, survivin, Bax and caspase-3 in experimental group D was significantly lower than that in other groups (P <0.05). The increase of Bcl-2 gene expression in experimental group D was also significantly higher than that in other groups (P <0.05). Conclusion Glycyrrhizin has a strong inhibitory effect on the proliferation of human breast cancer cell line MDA-MB-231 with significant dose-effect and time-dependent effects.
