目的:研究鸡骨草不同溶媒提取物对非酒精性脂肪肝(NAFLD)大鼠肝细胞固醇调节元件结合蛋白1c(SREBP-1c)表达的影响。方法:长期喂养高脂肪乳建立大鼠NAFLD模型,给予不同溶媒提取的鸡骨草溶液,检测大鼠血脂及肝组织总超氧化物歧化酶、丙二醛水平,并进行肝组织病理组织学检查及RT-PCR法检测SREBP-1c表达。结果:鸡骨草水提物可显著降低总胆固醇、甘油三酯、低密度脂蛋白胆固醇、丙二醛及SREBP-1c水平,显著升高高密度脂蛋白胆固醇、总超氧化物歧化酶水平;鸡骨草50%醇提物组的效果次之、95%醇提物组的效果不明显。结论:鸡骨草水提物能调节SREBP-1c水平,对NAFLD大鼠有一定治疗作用。
AIM: To investigate the effects of different extracts of Abrus cantharellese on the expression of steroid regulatory element-binding protein 1c (SREBP-1c) in nonalcoholic fatty liver disease (NAFLD) rats. Methods: The rat model of NAFLD was established by long-term feeding of high-fat milk. The rats were given the solution of Abrus cantoneiensis extracted by different solvents, the levels of serum lipids and total superoxide dismutase and malondialdehyde in liver tissue were detected, and histopathological examination SREBP-1c expression was detected by RT-PCR. Results: AstraZeneca extract could significantly reduce the levels of total cholesterol, triglyceride, low density lipoprotein cholesterol, malondialdehyde and SREBP-1c and significantly increase the levels of high density lipoprotein cholesterol and total superoxide dismutase. Crabgrass 50% alcohol extract group followed by the effect of 95% ethanol extract group effect is not obvious. Conclusion: Abrupt extract can modulate the level of SREBP-1c and have certain therapeutic effect on NAFLD rats.
