目的探讨花梨木精油改善小鼠抑郁样行为的可行性及其可能机制。方法将36只C57BL/6雄性小鼠,采用慢性不可预见性中等强度刺激(CUMS)结合孤养建立抑郁样小鼠模型,将筛选后的模型小鼠随机均分为CUMS+花梨木吸嗅组(花梨木组)、CUMS+盐酸氟西汀组(百忧解组)和CUMS组;另选12只小鼠为对照组。用神经行为学和免疫组化方法检测造模前后小鼠行为学及脑内多巴胺(DA)的变化。结果与CUMS组比较,花梨木组和百忧解组小鼠在吸嗅治疗2、3和4周时的悬尾安静和强迫游泳时间均明显缩短,而糖水消耗量明显增加(P<0.05)。花梨木组和百忧解组在海马、下丘脑、杏仁核DA等阳性反应产物较CUMS组多。结论花梨木精油吸嗅能够缓解抑郁症小鼠的抑郁样行为,其机制可能是通过嗅觉通路来调节脑的相应中枢而起作用。
Objective To investigate the feasibility and possible mechanism of rosewood essential oil to improve depression-like behavior in mice. Methods Thirty-six C57BL / 6 male mice were randomly divided into CUMS + rosewood aspiration group (control group) with CUMS + CUMS Rosewood group), CUMS + fluoxetine hydrochloride group (Prozac group) and CUMS group; another 12 mice were selected as the control group. Neurobehavioral and immunohistochemical methods were used to detect the changes of behavior and brain dopamine (DA) in mice before and after modeling. Results Compared with the CUMS group, the mice in the Huarui Mu group and the Prozac group had significantly decreased tail-tied quietness and forced swimming time at 2, 3 and 4 weeks of treatment, while the consumption of sugar and water significantly increased (P <0.05) . Rosewood group and Prozac group in the hippocampus, hypothalamus, amygdala and other positive reaction products DA more than CUMS group. Conclusion Rosin essential oil inhalation can alleviate depression-like behavior in depression mice, and its mechanism may be through the olfactory pathway to regulate the corresponding center of the brain to play a role.
