目的:考察牡丹皮提取物丹皮酚的体外黏膜渗透性及影响因素。方法:选择离体牛蛙皮肤为渗透模型,采用改良的Franz扩散池进行体外经皮吸收实验;HPLC测定丹皮酚的含量,研究接收液、蛙皮及药物浓度对丹皮酚体外黏膜渗透的影响。结果:接收液的渗透压和pH对丹皮酚的渗透有一定的影响;同一牛蛙不同部位的皮肤对丹皮酚渗透的影响较大,其表观渗透系数:腹皮>背皮>侧腹皮;丹皮酚黏膜渗透性良好,累积渗透率与时间符合一级吸收过程。结论:以离体牛蛙腹皮为渗透模型,丹皮酚的体外渗透性质与黏膜吸收的特点有相似性。
Objective: To investigate the in vitro mucosal permeability of Paeonia peel extract and its influencing factors. METHODS: The in vitro porcine skin was selected as the permeation model. The modified Franz diffusion cell was used for the in vitro percutaneous absorption test. The content of paeonol was determined by HPLC. The influence of the receiving liquid, frog skin and drug concentration on the in vitro mucosal penetration of paeonol was studied. . RESULTS: The osmotic pressure and pH of the receiving liquid had a certain influence on the penetration of paeonol; the skin of different parts of the same bullfrog had a greater influence on the penetration of paeonol, and its apparent permeability coefficient was as follows: abdominal skin> back skin> flanks Peel; paeonol mucous membrane permeability is good, cumulative permeability and time in line with the first-stage absorption process. Conclusion: The in vitro permeability profile of paeonol was similar to that of mucosal absorption in an in vitro model of the isolated bullfrog.
