目的:经多年实验研究,多糖具有体外及体内抗肿瘤作用,而体外抗肿瘤实验是抗肿瘤药物进行初步筛选的主要方法。我国苹果资源丰富,苹果粗多糖具有一定开发价值,从新鲜苹果中提取苹果粗多糖,并对其进行体外抗胃腺癌SGC7901肿瘤细胞增殖的研究。方法:采用醇沉法从新鲜苹果中提取粗多糖。新鲜苹果榨汁,取果汁经透析法除色素、Sevage法除蛋白、乙醇沉淀、冰冻干燥后得苹果粗多糖。采用MTF法观察苹果粗多糖不同作用时间、不同剂量对胃腺癌SGC7901肿瘤细胞增殖的抑制作用;采用集落形成实验法观察苹果粗多糖对胃腺癌SGC7901肿瘤细胞克隆形成率的影响。结果:获取苹果粗多糖,多糖获取率为0.1%。MTT法显示,苹果粗多糖(0.5,0.1,0.02mg/mL)在48h对体外胃腺癌SGC7901肿瘤细胞均有不同程度抑制作用(P<0.05),其中中浓度组和高浓度组的肿瘤抑制率分别达到10.8%和18.9%。在24h及72h抑制作用不明显。集落形成实验法显示,随着多糖给药量增加(0.02、0.1、0.5mg/mL)胃腺癌SGC7901肿瘤细胞集落形成率依次呈现97.10±2.33、95.30±3.62、93.6±5.66的下降趋势。结论:苹果粗多糖对胃腺癌SGC7901肿瘤细胞有较强的体外抗肿瘤作用,并呈现一定的量效关系。
OBJECTIVE: After years of experimental research, polysaccharides have antitumor activity in vitro and in vivo, while in vitro antitumor experiment is the primary method for primary screening of antitumor drugs. Apple is rich in resources in our country. Apples crude polysaccharides have some development value. Apple crude polysaccharides are extracted from fresh apples and the proliferation of SGC7901 cells in vitro is studied. Methods: Crude polysaccharides were extracted from fresh apples by alcohol precipitation method. Fresh apple juice, take juice by dialysis method in addition to pigment, Sevage France protein, ethanol precipitation, freeze-dried apple polysaccharides. MTT assay was used to observe the inhibitory effect of different concentrations of crude polysaccharides on the proliferation of SGC7901 gastric cancer cells. The colony formation assay was used to observe the effect of crude polysaccharides on the colony formation of gastric adenocarcinoma SGC7901 cells. Results: Obtained apple crude polysaccharides, polysaccharide access rate of 0.1%. MTT assay showed that the crude polysaccharides (0.5,0.1,0.02mg / mL) inhibited SGC7901 cells in vitro to a certain extent (P <0.05) at 48h, and the tumor inhibition rates of medium and high concentration groups Reaching 10.8% and 18.9% respectively. 24h and 72h inhibition is not obvious. Colony formation assay showed that the colony formation rate of gastric adenocarcinoma SGC7901 cells showed a decreasing trend of 97.10 ± 2.33, 95.30 ± 3.62 and 93.6 ± 5.66 with the increase of polysaccharide dosage (0.02,0.1,0.5mg / mL). Conclusion: The crude polysaccharides of apple have stronger anti-tumor effect on gastric adenocarcinoma SGC7901 tumor cells in vitro, and show a certain dose-effect relationship.
